酸奶在制作过程中要进行高温杀菌，目前乳品厂多采用巴氏杀菌，（上海冷库安装）即采用95℃的高温进行杀菌处理。酸奶灭菌之后，还要进行高温发酵（发酵温度在40℃~43℃之间），发酵之后便要采取冷却措施。冷却措施就是让酸奶发酵后快速冷却到5℃，因为酸奶中含有的保加利亚乳杆菌和噬热链球菌，它们的生长温度分别为22℃和20℃。如果在规定时间内达不到温度要求，则会严重影响酸奶的菌群数量、营养物质、风味等。
